Application Notes
Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. In human PBMC, a ~120 kDa band is observed. A smaller isoform, TLR9 isoform B (Genbank accession no. AAF72190) containing 975 amino acids may also be observed in some cases. In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.

Background

Toll-like receptors (TLRs) are evolutionarily conserved pattern-recognition molecules resembling the toll proteins that mediate antimicrobial responses in Drosophila. These proteins recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. TLR9 forms a subfamily along with TLR7 and TLR8 that recognize viral RNA and CpG DNA sequences and are localized in intracellular acidic compartments such as the phagolysosome. Unlike other TLRs which act through adaptor molecules such as TOLLIP, TIRAP, TRIF, and MyD88 to activate various kinases and transcription factors to respond to potential infection, TLR9 is strictly dependent on MyD88.

  1. I am a bit unsure about your antibody NBP2-24729 specific against TLR9. I used it for  probing mouse B cell lysate on Proteisimple WES. On their website the recommend your antibody and claim that it detects a specific signal at 90 kDa. But on your website simple western detects a band at 200 kDa?
    • This came during the race to 1000 and we had many people generating and reviewing data.  In retrospect, this was one that probably should not have made it out. As I reviewed the run data, it looks as though the MW ladder did not assign correctly.  When this was corrected, the band at 200 actually shifts higher, up to 280 kDa.  This band now overlaps with the 280 kDa fluorescent standard which is prone to have non-specific interactions with some antibodies.  Those two things make this non-supportive in Simple Western.   We ran Raji lysate on the same run.  This antibody detects a very broad band with a peak at ~150-165 kDa, closer to expected MW.  However, the baseline for this samples was also very high.  If we were presented with this data set today, it would not be considered supportive.  The other information on the Protein Simple web site likely came from a customer.
