This USP9x antibody is useful for Immunocytochemistry/Immunofluorescence, Immunohistochemistry paraffin embedded sections and Western blot, where a band is seen ~270 kDa. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Background

USP9x (ubiquitin-specific protease 9, X chromosome) is one of the largest deubiquitinases (DUBs) belonging to the Ub-specific protease family and implicates in processing of ubiquitin precursors as well as of ubiquitinated proteins, thereby playing a key regulatory role at protein turnover level by preventing degradation of proteins through conjugated ubiquitin removal. USP9X substrates include AF-6, beta-catenin, NUAK1, MARK4, ErbB2, EFA6, Smad4, Mcl1, ASK1, and survivin, and USP9X has been involved in several biological processes that include: protein trafficking and tight junction biogenesis in epithelial cells; regulation of the TGF-beta pathway; chromosome alignment and segregation; component of PEX5-mediated peroxisomal protein import pathway. Moreover, USP9X modulates the proteins levels of: alpha-synuclein, a protein involved in the pathogenesis of Parkinson disease; MCL1, an anti-apoptotic protein generally found overexpressed in cancers; some protein kinases involved in stress responses and cellular energy homeostasis; and some ubiquitin-ligases, such as MARCH7 and Itch.
