Application Notes
In Western blot, this antibody detects a single band at 37 kDa. In IHC, it can be competitively inhibited from recognizing the APE1 antigen in tissues using APE1 protein. It can also be used on frozen and fixed-paraffin sections and cytospin preps. In IHC-P, staining was observed in the nucleus of a human breast cancer xenograft. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.

Background

The mammalian apurinic/apyrimidinic endonuclease (APE/ref-1) is responsible for the repair of AP sites in DNA. In addition, this enzyme functions as a redox factor facilitating the DNA binding capability of FOS, JUN, NfkB, HIF-1 alpha, Pax-5, Pax-8 and other transcription factors. APE/ref-1 has also been shown to control p53 activity through redox alteration. APE is linked to apoptosis, associated with thioredoxin, and altered levels of APE/ref-1 have been found in some cancers. APE appears to form a unique link between the DNA base excision pathway, oxidative signaling, transcription regulation, cancer and cell-cycle control.

  1. I have problems performing IF on HeLa cells with this antibody (NB100-116). I have a very weird pattern of staining. Is there a special protocol I need to follow?
    • Please see the following link for our product specific protocol for NB100-116.
  2. Today I have a customer who is having some difficulty with NB100-116 in his IP experiments. I have sent him a link to a journal that references use of the antibody in IP, but he has now asked if the antibody recognizes the reduced form of APE1 and not the oxidized form. Do you have any information available?
    • It seems that in Western blot, Novus as well as our customers use reducing conditions, as is standard. I have also looked at our publications using this antibody in IP and nothing suggests that it needs to be used in reducing conditions for IP. It would seem that it recognizes both forms and should work in IP in most standard IP protocols.
