Background

Factor Inhibiting HIF1 (FIH) protein facilitates repression of HIF-1 transcriptional activity by binding to VHL, which acts as a transcriptional corepressor. VHL inhibits HIF-1 alpha transactivation function by recruiting histone deacetylases. Involvement of VHL in association with FIH provides a unifying mechanism for the modulation of HIF-1 alpha protein stabilization and transcriptional activation in response to changes in cellular oxygen concentration.

Some cells remained at normoxia for the duration of the culture (N); others were at normoxia for 35 h and then hypoxia (1% O2) for 24 h (H).
