Background
Anabolic signals such as glucose, insulin etc. enhance the transcription of lipogenic enzymes responsible for converting excessive carbohydrates into triglycerides, whereas, DGAT1 (diacylglycerol O-acyltransferase 1) is an enzyme that facilitates the conversion of fatty acids to triglycerides. DGAT1 is a multi-pass membrane protein localized in endoplasmic reticulum which catalyzes the terminal and only committed step in triacylglycerol synthesis by using diacylglycerol and fatty acyl CoA as substrates. In contrast to DGAT2, it is not essential for survival, and is involved in VLDL assembly. In liver, DGAT1 plays a key role in esterifying exogenous fatty acids to glycerol, and is important in higher eukaryotes for physiologic processes involving triacylglycerol metabolism such as intestinal fat absorption, lipoprotein assembly, adipose tissue formation, and lactation. Moreover, it acts as the major acyl-CoA retinol acyltransferase (ARAT) in skin tissues, wherein, it maintains the retinoid homeostasis for preventing retinoid toxicity responsible for skin and hair disorders.
