Transcription factor PU.1 belongs to the ETS family and contains one ETS DNA-binding domain. It is a novel caspase-3 substrate and found to contribute partially chemotherapeutic agents-induced apoptosis in leukemic cellsThis protein is also reported to activate gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. It is found to be one of the key regulators of hematopoitic cell development and induces apoptosis in myeloma cells through direct transactivation of an apoptosis-related gene, tumor necrosis factor (TNF)-related apoptosis inducing ligand (TRAIL).
