Western Blot: PU.1/Spi-1 Antibody (PD00-14) [NBP3-32878] - Western blot analysis of PU.1/Spi-1 on different lysates with Rabbit anti-PU.1/Spi-1 antibody (NBP3-32878) at 1/500 dilution. Lane 1: Daudi cell lysate Lane 2: THP-1 cell lysate Lysates/proteins at 10 ug/Lane. Predicted band size: 31 kDa Observed band size: 40 kDa Exposure time: 2 minutes; 12% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32878) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Background

PU.1/Spi1 encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]
