Specificity
Specific for human DAZL, a member of the DAZ (deleted in azoospermia) family of RNA binding proteins. DAZL is expressed in fetal and adult testes and ovaries, and is believed to play a role in germ cell development. In adult germ cells, the expression of DAZL is localized to the cytoplasm. Mutations in this gene have been linked to severe spermatogenic failure and infertility in males. Clone DAZL/4253 recognizes DAZL at its C terminus, and is used in detection of DAZL expression by various methods.

Background

DAZL is a RNA-binding protein, which is essential for gametogenesis. Plays a central role during spermatogenesis. May act by binding to the 3'-UTR of mRNA and thereby regulating the translation of key transcripts

BG01V human embryonic stem cells were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. Nanog/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human Nanog Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1997) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The nanog promoter was detected by standard PCR.
