Crk belongs to a family of adaptor molecules that are widely expressed in numerous tissues. It was originally isolated from a chicken tumor sample, and despite lacking enzymatic activity, this protein is involved in many signal transduction pathways leading to phosphorylation of multiple proteins. Crk forms multi protein complexes via it’s SH2 (aa 13-118) and SH3 (aa 132-192 and aa 237-296) domains leading to activation of downstream molecules involved in growth, differentiation, cell adhesion and migration. Alternate splicing generates an additional isoform that lacks aa 205-304.
