Immunocytochemistry/Immunofluorescence: B7-H2/ICOSLG Antibody (HK5.3) - Chimeric [NBP3-09018] - Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) splenocytes on Shi-fix(TM) coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of HK5.3 (NBP3-09018) at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (1 ug/ml), showing weak membrane staining in a subset of cells. Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2a format, for improved compatibility with existing reagents, assays and techniques.

This antibody has been used to detect murine ICOSL by flow cytometry. It is able to block ICOSL interacting with ICOSL and inhibit T-cell proliferation in a collagen type II (CII)-induced arthritis model.

Background

B7-RP1 is a 40 kD protein, also known as ICOS ligand, B7-H2, and B7h. It has recently been clustered as CD275. It is a member of the immunoglobulin receptor superfamily expressed on splenic B cells, dendritic cells and macrophages. The expression of this antigen is increased by TNF- alpha stimulation and is highly upregulated by local inflammation. B7RP1 binds to ICOS and co-stimulates T cell and B cell responses. Two isoforms of this protein have been reported to occur as a result of alternative splicing. These isoforms show differential tissue distribution.
