Background
The mammalian transient receptor potential (TRP) superfamily can be divided into three major families including the "canonical TRP" (TRPC) family. The seven members of this family share the activation through PLC-coupled receptors and have been suggested to be components of receptor-regulated cation channels in different cell types. Furthermore, the members of the TRPC6/6/7 subfamily can be activated by diacylglycerol (DAG) analogs, suggesting a possible mechanism of activation of these channels by PLC-coupled receptors. When expressed in transfected cells, TRPC6 acts as a non-selective store-independent receptor-activated cation channel. It is activated by DAG in a PKC-independent manner and is insensitive to IP3 activation. There is increasing evidence that TRPC6 encodes endogenous DAG-activated receptor-operated cation channels in vivo.

I have tested this antibody and would like to share the results from my first preliminary experiment. This Ab has been tested in ICC using a human cell line. Blocking buffer contains either goat or donkey serum. Dilutions: both 1:1000 and 1500 have been tested. Secondary ab is goat anti rabbit FITC (1:400). Results: every single cell shows VERY WEAK signal around only the cell body. No signal was detected in any cytoplasmic processes, which is really different from TRPM2. As the signal is weak, I will increase the concentration by using the following dilutions instead: 1: 200 and 1:500 to find out if the staining is getting stronger. Questions: Despite the weak staining, every single cell has been stained. Thus, (1) How can I confirm that the staining is specific? (2) How can confirm the staining is in the cell membrane?
- Membrane staining should be around the edge of the cell. If this is not what you are seeing then I would suspect that your staining is unsuccessful. You may need to permeabilize since this antibody is expected to stain the intracellular portion of TRPC6 and what you're seeing may simply be background and non-specific. Once again, confirmation of staining should best be discussed with your mentor who should have greater knowledge on your proteins of interest than I do.
