p73 Antibody (5B429) - BSA Free Summary

Immunogen
This antibody was raised against full-length human p73. It reacts with an epitope that is located on the N-terminal region of human p73. It does not cross react with p53.
Specificity
p73 antibody (clone 5B429) recognizes all of the TAp73 isoforms i.e. isoforms with an intact NH2-terminal transactivation domain (Thottassery et el, 2006, PMID 16505115). Sayan et al 2005 (PMID 15781249) showed that p73 IMG-246/ NBP2-24737 reacted with all tested TAp73 isoforms but not deltaNp73 whereas p73 antibody clone 5B1288 (NB100-56674 / IMG-259A ) showed reactivity against all p73 isoforms including delta Np73. Caspase-cleaved p73 fragments, including 55 kDa and 45 kDa, have been described in literature (Sayan et al, 2008).

Background

p73 is a transcription factor that shares a significant amino acid identity with p53 and p63 (1). p73 and p63 can both activate p53-responsive genes and induce apoptosis upon over expression (2) Unlike p53, p73 is not induced by DNA damage and is not a target for inactivation by viral oncoproteins (3). Similar to p63, p73 has an additional coding region in the C-terminus (not found in p53) that can undergo complex alternative splicing giving rise to 3 splice forms (alpha, beta, and gamma) (4). Activation of p73 requires the presence of functional c-Abl. It has also be found that mutant p53 can bind to and inactivate p73 (5).

  1. Do you know if it also detects deltaN isoforms?
    • For product NPB2-24737, the immunogen was the full length human p73 protein isoform alpha. The epitope for this product has been mapped to the N-terminal region of this isoform. This region shares 100% homology with isoform delta, so it should also recognize this isoform as well.
  2. Also, have you guys mapped the epitope?
    • While we know the general region where the epitope lies, we have not yet mapped the exact epitope.
