Background
DDX3 is a conserved RNA helicase ubiquitously expressed in the cytoplasm. A 73kD protein and member of the DEAD-box family, DDX3 appears to have important regulatory activities in a variety of cell programs including growth and cell cycle control, apoptosis, and anti-viral signaling pathways. HIV and HCV co-opt the DDX3 molecule as an essential host factor for their replication. Additionally DDX3 appears to recognize and sense viral RNA, interacts with IPS-1 dependent signaling pathway molecules, and TRIF, an adaptor molecule in TLR3 and TLR4 signaling pathways, as well as ZBP1 which also is involved in upregulation of IFNb by HCMV. Loss of DDX3 may lead to enhanced proliferation and anti-apoptotic programs -pathways that promote a role for DDX3 in tumorigenesis. The availability of antibody specific for DDX3 provides opportunity to further detail its potential role in anti-viral immunity as well as serving as a novel target for intervention.
