Background
Cadherins are a family of transmembrane adhesion receptors that are involved in the formation of cell to cell junctions in a variety of tissues, which is crucial for the development, maintenance and homeostasis of tissue architecture and function. Cadherin expression is regulated spatially as well as temporally. The cadherin superfamily of proteins contains over 100 members. Classical cadherins (e.g., N-, E-, and C-cadherin) consist of five CADs, a single transmembrane helix, and a highly conserved cytosolic domain (koch AW et al., 2004). These cadherins feature a 24-amino acid sequence in the carboxy terminal which is conserved among cadherin family proteins and across animal species; antibody generated against this sequence are termed pan-cadherin antibody. Cadherins depend on calcium for their function: removal of calcium abolishes adhesive activity and renders cadherins vunerable to proteases. The cytoplasmic domain of classical cadherins interacts with beta-catenin, gamma-catenin (also called plakoglobin), and p120 catenin. Ctnnb and Ctnng associate with Ctnna, which links the cadherin-catenin complex to the actin cytoskeleton. Deletion of the cytoplasmic domain destroys these interactions and also eliminates cadherin function. Recently Src, Yes, and Lyn gene products of the Src proto-oncogene family have been found to be responsible for Cadherin phosphorylation (Shapiro L et al., 2009). Cadherins are differentially expressed during development and in adult organs. Since many cell types express multiple Cadherin subclasses simultaneously, it can be inferred that the adhesion properties of individual cells are thus governed by varying the combinations of cadherins. There is also recent evidence to suggest that altered expression of cadherins may be involved in invasion and metastasis of tumor cells (Maret D et al., 2010).
