Specificity |
It recognizes an oncofetal glycoprotein with a single chain of 70kDa, which is identified as alpha-fetoprotein (AFP) (ISOBM TD-2 workshop). This monoclonal antibody is highly specific to AFP and shows no cross-reaction with other oncofetal antigens or serum albumin. AFP is normally synthesized in the liver, intestinal tract, and yolk sac of the fetus. Antibody to AFP has been shown to be useful in detecting hepatocellular carcinomas (HCC) and germ cell neoplasms, especially yolk sac tumors. |

Background
Alpha 1 Fetoprotein encodes alpha-fetoprotein, a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha-fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha-fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha-fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha-fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly.

How do you purify product Cat# NBP2-34577AF700 after conjugation? As the datasheet says, this product is Protein A purified, I would like to know whether the antibody was purified with Protein A before or after conjugation.
- The antibody is Protein A purified before conjugation. After the conjugation, for all small dyes like AF700 or AF647, we only use a small desalting column to remove excess of the dye.
