Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-ZEB1 antibody NBP1-05987 used for IP at 3 ug per reaction. ZEB1 was also immunoprecipitated by another rabbit anti-ZEB1 antibody. For blotting immunoprecipitated ZEB1, NBP1-05987 was used at 0.4 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Immunocytochemistry/Immunofluorescence: PD-L1 Antibody [NBP1-76769] - U-251 MG cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NBP1-76769 at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: PD-L1 Antibody [NBP1-76769] - An intracellular stain was performed on U-251 MG cells with PD-L1 Antibody NBP1-76769 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Dual RNAscope ISH-IHC: PD-L1 Antibody [NBP1-76769] - PDCD1 mRNA (red) and PD-L1 protein (green) were detected in formalin-fixed paraffin-embedded tissue sections of human lung cancer. ACD's Integrated Co-Detection Workflow was performed using ACD RNAScope Probe Hs-PDCD1 and PD-L1 antibody at 1:100 dilution. Tissue was stained on Leica Bond RX using RNAscope (TM) 2.5 LS Reagent Kit-RED, BOND Polymer Refine Detection (DAB) and Hematoxylin, BOND Polymer Refine Red Detection and Hematoxylin and RNAscope (TM) 2.5 LS Green Accessory Pack. Tissue was counterstained with 50% hematoxylin (blue).

Immunocytochemistry/Immunofluorescence: LRRK2 Antibody [NB300-268] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-LRRK2 Antibody NB300-268 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: LRRK2 Antibody [NB300-268] - An intracellular stain was performed on U2OS cells with LRRK2 Antibody NB300-268 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: LRRK2 Antibody [NB300-268] - LRRK2 antibody was tested in SH-SY-5Y cells at a 1:200 dilution against DyLight 488 (Green). Alpha tubulin and nuclei were counterstained against DyLight 568 (Red), and DAPI (Blue), respectively.

BRD4770-induced activation of the FA pathway does not occur via the direct induction of DNA damage or altered cell cycle progression(A) BJ-TERT cells were incubated with (+) and without (-) 200 nM mitomycin C (MMC) in the absence (NT) or presence of 10 μM BRD4770 for 24 h. Knockout Validated: FUS Antibody [NB100-565] - Western blot shows lysates of HEK293 human embryonic kidney parental cell line and FUS knockout (KO) HEK293 human embryonic kidney cell line. PVDF membrane was probed with 1:1000 of Rabbit Anti-Human FUS Polyclonal Antibody (Catalog # NB100-565) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for FUS at approximately 75 kDa (as indicated) in the parental HEK293 celll line, but is not detectable in the knockout HEK293 cell line. This experiment was conducted under reducing conditions.

<b>Genetic Strategies Validation. </b>Western Blot: FUS Antibody [NB100-565] - FUS activates GSK-3beta in transfected cells and transgenic mice A. Cells were transfected with either control vector (CTRL), HA-FUS, HA-FUSR521C or HA FUSR518K and the samples probed on immunoblots for GSK-3beta phosphorylated on serine 9 (GSK-3beta-S9), total GSK-3beta, FUS (using FUS antibody) and tubulin as a loading control. Phosphorylation of GSK-3beta serine 9 is the principal mechanism for regulating its activity; serine 9 phosphorylation inhibits GSK-3beta activity. <b>Genetic Strategies Validation. </b>Knockdown Validated: Rictor Antibody [NB100-612] - SERCA2A expression is regulated by mTORC2. Rictor silencing was induced by cell transfection with rictor siRNA and then, cells were stimulated with IGF-1 for 24 hours in presence or absence of E2. mTORC2 downregulation was confirmed by abolished Akt-pS473 and resulted in decreased SERCA2A protein expression. Akt-pT308, mTOR and raptor were not negatively affected by rictor silencing. Pictured are representative western blots. Antibodies: Affinity purified rabbit anti-Rictor antibody NB100-612 (lot NB100-612-4) used for IP at 3 ug per reaction. Rictor was also immunoprecipitated by a previous lot of this antibody (lot NB100-612-3) and rabbit anti-Rictor antibody NB100-611. For blotting immunoprecipitated Rictor, NB100-612 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Western Blot: Rictor Antibody [NB100-612] - mTOR complex protein expressions & rictor phosphorylation at Ser1235. Cardiomyocytes were pretreated for 30 min with 20 nM rapamycin, then stimulated with 10 nM IGF-1 in presence/absence of 10 nM E2 for 24 h. Western blots for GSK3beta-pS9, GSK3beta, rictor pS1235 and rictor. At least 3 independent experiments indicate that rictor phosphorylation at S1235 by GSK-3beta downregulates mTORC2 activity. IGF-1 induced strong phosphorylation of GSK-3beta at Ser9 in the absence and presence of E2. Rapamycin pretreatment reduced phosphorylation in E2 co-treated cardiomyocytes, indicating increased activity of GSK-3beta, which was not associated with increased phosphorylation of rictor at S1235. PHF8 is upregulated by HIFs during hypoxia and regulates histone methylation in response to hypoxia. (a–c) LNCaP (a), 22RV1 (b) and DU145 (c) cells were cultured first at normoxic conditions and then shifted to hypoxic conditions for 0, 12, 24 h. Cells were collected and whole-cell lysates were prepared. Proteins were separated on 8% SDS–polyacrylamide gel electrophoresis and western blotting was performed with antibodies against HIF1 alpha , HIF2 alpha , PHF8, GLUT-1 and beta -actin as indicated. The total RNAs were then prepared and quantitative RT–PCR for PHF8 was performed. The results are presented as the mean±s.e.m. of three independent experiments. (g) Knockdown HIF2 alpha or HIF1 alpha abrogated hypoxia-induced upregulation of PHF8 proteins. LNCaP cells stably infected with control shRNA (shCtrl) or shHIF2 alpha (left panel) /shHIF1 alpha (right panel) were cultured under normoxic (N) or hypoxic (H) conditions for 12 h and then collected for western blot analysis using antibodies as indicated. Immunolocalization of HIF1A in limbs.Immunoreactivity was detected in control limbs in the apical ectodermal ridge and interdigital regions (I.R.). The mesenchymal condensations/developing cartilaginous anlagen (Digits) were not immunoreactive. 4-OOHCPA treatment resulted in a concentration- and time-dependent increase in HIF1A immunoreactivity in this area in the 3 and 6 h exposure groups; staining was diminished by 24 h.
