Background
TRAF3IP1, or MIP-T3 functions as a down regulator of type I IFN responses, a critical component of the innate immune defense in viral infection (1, 2). TRAF3IP1 is involved in the suppression of IL-13 and is effected through IL-13 induction of STAT6 phosphorylation (1). TRAF3IP1 interacts and inhibits TRAF3 complex formation and subsequent downstream signaling molecules RIG-I, MDA5 RIG-I like innate immune receptor molecules, VISA and MAVS, a pathway which leads to IRF mediated IFN-g responses. Additionally TRAF3IP1 also interferes with the signaling through TBK1 and IKKe a common juncture between RLR and TLR signaling (3, 4). It is TRAF3 interaction and binding with MAVS and inhibition of this complex in RLR signaling pathways as well as TRAF3IP1 regulation of TLR signaling that highlight a potential new target for studying IFN responses and regulating them. Antibody to TRAF3IP1 provides a unique tool to further development of regulators of IFN production.
