CARD9 (caspase recruitment domain-containing protein 9) is a 70 kDa member of the CARD module family of proteins. It is expressed in myeloid lineage cells such as macrophages, neutrophils and dendritic cells and mediates activation of p38 and Jnk, as well as NF-kappa B by interacting with BCL10 and MALT1 in a PKC-independent manner. Human CARD9 is 536 amino acids (aa) in length. It contains one CARD domain (aa 6‑98), two coiled-coil regions (aa 117‑277 and 332‑419) and a phosphorylation site at Ser460. CARD9 dimerizes with itself and BCL10. Multiple potential splice variants are noted for CARD9. There are 35 aa and 39 aa substitutions for aa 479‑536, a seven aa substitution for aa 360‑536 and an eleven aa substitution for aa 482‑536. Over aa 9‑256, human CARD9 is 88% aa identical to mouse CARD9.
