Western blot shows lysates of bEnd.3 mouse endothelioma cell line. PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human/Mouse/Rat CD31/PECAM-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3628) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). A specific band was detected for CD31/PECAM-1 at approximately 130 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Rat splenocytes were stained with Goat Anti-Human/Mouse/Rat CD31/PECAM-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3628, filled histogram) or isotype control antibody (AB-108-C, open histogram), followed by Allophycocyanin-conjugated Anti-Goat IgG Secondary Antibody (F0108).

CD31/PECAM-1 was detected in immersion fixed bEnd.3 mouse endothelioma cell line using Goat Anti-Human/Mouse/Rat CD31/PECAM-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3628) at 10 µg/mL for 3 hours at room temperature. Tissue was stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (yellow; NL001) and counterstained with DAPI (blue). CD31/PECAM-1 was detected in immersion fixed frozen sections of mouse embryo (14 d.p.c.) using Goat Anti-Human/Mouse/Rat CD31/PECAM-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3628) at 10 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (VC004). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to endothelial cells in vasculature.

AcSDKP suppresses TGF beta /smad signaling and EndMT through the FGFR1/FRS2 pathway. Diminished permeability of liver sinusoids in Plvap-deficient mice.Neither by immunohistochemistry with antibodies against CD31 (A) nor by light microscopy of 1 µm semi-thin sections (B, Richardson's stain) obvious differences are detected with regards to the overall orientation and the density of liver sinusoids between 3-week-old Plvap-/- mice and wild-type littermates. Sinusoids of Plvap-deficient mice show a higher number of macrophages in their lumen (white arrows) and focal areas with accumulations of mononuclear cells in Disse's space (black arrows). PVDF membrane was probed with 1 µg/mL of Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for MMR/CD206 at approximately 180 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

MMR/CD206 was detected in perfusion fixed frozen sections of mouse testis using Goat Anti-Mouse MMR/CD206 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2535) at 5 µg/mL overnight at 4 °C. M1 and M2 phenotype in spinal cord after intraplantar IL-1 beta . Wild-type (WT) and LysM-G protein–coupled receptor kinase (GRK)2+/− mice received an intraplantar injection of 1 ng IL-1 beta . At 15 hours after injection, spinal cord was collected, and frozen sections of (A) lumbar spinal cord (L2 to L5) and as control (B) thoracic spinal cord (T6 to T10) were stained for M1 (CD16/32) and M2 (CD206 and arginase-I) phenotypic markers. Western blot shows lysates of HEK293T human embryonic kidney cell line either mock transfected or transfected with full length mouse IL-33. PVDF membrane was probed with 0.4 µg/mL of Goat Anti-Mouse IL-33 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3626) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for IL-33 at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

IL-33 was detected in immersion fixed frozen sections of mouse spleen using Goat Anti-Mouse IL-33 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3626) at 5 µg/mL overnight at 4 °C. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red, upper panel; Catalog # NL001) and counterstained with DAPI (blue, lower panel). Mouse splenocytes were treated for 5 hours with 50 ng/mL PMA and 1 μg/mL Ca2+ ionomycin then stained with Goat Anti-Mouse IL‑33 Antigen Affinity‑purified Polyclonal Antibody (Catalog # AF3626, filled histogram) or isotype control antibody (Catalog # AB-108-C, open histogram), followed by Phycoerythrin-conjugated Anti-Goat IgG Secondary Antibody (Catalog # F0107). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.

Recombinant Mouse IL‑33 (Catalog #3626-ML) stimulates proliferation in the D10.G4.1 mouse helper T cell line in a dose-dependent manner (orange line), as measured by Resazurin (Catalog # AR002). Proliferation elicited by Recombinant Mouse IL‑33 (0.25 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Mouse IL‑33 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3626). The ND50 is typically 10-50 ng/mL.

Western blot shows conditioned media from CHO Chinese hamster ovary cell line either mock transfected or transfected with human TNF- alpha. PVDF membrane was probed with 2 µg/mL of Goat Anti-Human/Mouse TNF-a Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-410-NA) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). Specific bands were detected for TNF-a at approximately 14 and 17 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Recombinant Mouse TNF-a (410-MT) induces cytotoxicity in the the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line). Cytotoxicity elicited by Recombinant Mouse TNF-a (0.1 ng/mL) is neutralized (green line) by increasing concentrations of Human/Mouse TNF-a Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-410-NA). The ND50 is typically 1.5-10 ng/mL in the presence of the metabolic inhibitor actinomycin D.

ANXA8+ve/c-kit+ve luminal progenitor cells are mostly Ki67−ve.Co-immunofluorescence staining for ANXA8 (green), c-kit (red), and Ki67 (purple) in mouse mammary gland from virgin and mid-pregnant mice shows that ANXA8+ve cells express c-kit, but not Ki67. VEGFR2/KDR/Flk-1 was detected in immersion fixed frozen sections of mouse embryo (14 d.p.c.) using 15 µg/mL Goat Anti-Mouse VEGFR2/KDR/Flk-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF644) overnight at 4 °C. Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Specific labeling was localized to mesenchymal cells. VEGFR2 was detected in acetone fixed cryosections of mouse kidney tissue using Goat Anti-Mouse VEGFR2/KDR/Flk-1 Polyclonal Antibody (Catalog # AF644) for 50 minutes at room temperature. Tissues were stained with rabbit anti-goat secondaryantibody and HRP polymer-conjugated anti-rabbit IgG followed by AEC+Substrate Chromogen (red) followed by counterstaining with hematoxylin (blue). Experiments were carried out and the image was provided by Dr. Grietje Molema, University Medical Center Groningen, The Netherlands.

Recombinant Mouse VEGFR2/KDR/Flk‑1 Fc Chimera (Catalog # 443-KD) inhibits Recombinant Mouse VEGF164 (Catalog # 493-MV) induced proliferation in HUVEC human umbilical vein endothelial cells in a dose-dependent manner (orange line). Inhibition of Recombinant Mouse VEGF164 (5 ng/mL) activity elicited by Recombinant Mouse VEGFR2/KDR/Flk‑1 Fc Chimera (50 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Mouse VEGFR2/KDR/Flk‑1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF644). Western blot shows lysates of human primary naïve CD4+ T cells, along with whole cell lysates (WCL) and conditioned-media supernatant (Supe) of human primary differentiated Th17 cells. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human IL-17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). A specific band was detected for IL-17 at approximately 15 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

IL‑17/IL‑17A was detected in immersion fixed paraffin-embedded sections of human tonsil using Goat Anti-Human IL‑17/IL‑17A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA) at 1 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes. Human IL-17 was immunoprecipitated from 100 µg of human primary differentiated Th17 cell lysate following incubation with 3 µg Goat Anti-Human IL-17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA) or control antibody (AB-108-C) overnight at 4 °C. IL-17-antibody complexes were absorbed using Protein G Sepharose. Immunoprecipitated IL-17 was detected by Western blot using 1 µg/mL Goat Anti-Human IL-17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA). Human peripheral blood mononuclear cells were unstimulated (light orange filled histogram) or treated with 50 ng/mL PMA and 250 ng/mL Ca2+ ionomycin for 16 hours, then stained with Goat Anti-Human IL‑17 Antigen Affinity‑purified Polyclonal Antibody (Catalog # AF‑317‑NA, dark orange filled histogram) or isotype control antibody (Catalog # AB-108-C, open histogram), followed by Allophycocyanin-conjugated Anti-Goat IgG Secondary Antibody (Catalog # F0108). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.

Recombinant Human IL‑17 (Catalog # 317-ILB) stimulates IL‑6 secretion in NIH/3T3 mouse embryonic fibroblasts in a dose-dependent manner (orange line), as measured by the mouse IL‑6 Quantikine ELISA Kit (Catalog # M6000B). IL‑6 secretion elicited by Recombinant Human IL‑17 (15 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human IL‑17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA). The ND50 is typically0.02‑0.12 µg/mL.

Expression of psoriasis‐related cytokines in paradoxical psoriasiform reactions. Immunohistochemistry analysis performed on paradoxical skin lesions obtained from patients 1 (Pt1) and 2 (Pt2) shows similar values of IL‐17A+ cells, a reduction of dermal IFN‐ gamma + cells and an increase of IL‐22+ or IL‐36 gamma + cells, when compared with psoriatic skin lesions. LS and NLS skin of the same psoriatic patient (n = 3) was analyzed. Graphs show the mean of number of positive cells + SD per three sections.
