Background
VG5Q (vasculogenesis gene on 5q protein, also called AGGF1) is a novel angiogenic factor that has been identified through genetic analysis of Klippel-Trenaunay syndrome (KTS), a congenital vascular disorder composed of capillary malformations, venous malformations or varicose veins, and hypertrophy of the affected tissues. VG5Q is broadly expressed in different cell types including endothelial cells (umbilical vein endothelial cells and microvascular endothelial cells), vascular smooth muscle cells and osteoblasts. VG5Q expression is mainly cytoplasmic and upon angiogenic stimulation especially when endothelial cell tube formation is initiated, VG5Q gets secreted and interact with a secreted angiogenic factor TNFSF12. VG5Q promotes angiogenesis as well as the proliferation of endothelial cells through direct binding in an autocrine fashion. Moreover, it has been demonstrated to promote angiogenesis as potentially as VEGF and its expression decreases with knockdown of GATA1 gene which is proposed as VG5Q regulator through two GATA1 binding sites in VG5Q promoter region.
