Background
Striatin (STRN) is a calmodulin-binding member of WD-repeat family of proteins/WD repeat striatin family and is an intracellular protein localized mainly in cytoplasm as well as in cellular membranes as peripheral membrane protein, and is mostly present in neurons of mammalian basal ganglia and cranial/spinal motor nuclei. Striatin's protein-protein interaction domains include caveolin-binding (amino acids 55-63), coiled-coil (amino acids 70-116), Ca2+-calmodulin-binding (amino acids 149-166), and a series of eight WD repeat (amino acids 419-780) motifs. Striatin is a scaffolding protein which functions in a calcium dependent fashion and is implicated in cellular signaling and trafficking processes. Striatin forms complex with PP2A (protein phosphatase 2A) which associates directly to and controls the function of ER-alpha. Moreover, striatin targets ER-alpha to the cell membrane and serves as a scaffold for the assembly of proteins essential to rapid, estrogen-induced eNOS activation. Striatin has also been suggested as a candidate gene for human ARVC (arrhythmogenic right ventricular cardiomyopathy), familial cardiac disease characterized by ventricular arrhythmias and sudden cardiac death.
