Background
SF3B4 (splicing factor 3B subunit 4, also known as SF3b50 or SAP49), is a ubiquitously expressed nuclear protein which forms a subunit of splicing factor SF3B (SF3B1/SAP155/SF3B155, SF3B2/SAP145/SF3B145, SF3B3/SAP130/SF3B130, SF3B4/SAP49/SF3B49, SF3B14A, PHF5A/SF3B14B, SF3B10 and SF3B125) required for 'A' complex assembly formed by the stable binding of U2 snRNP to pre-mRNA's BPS (branchpoint sequence). SF3B4 interacts directly with SF3B2 and also forms a component of U11/U12 snRNPs that are a part of the U12-type spliceosome. SF3B4 has also been suggested to implicate in 'E' complex assembly and is also found in 'B' and 'C' complexes. SF3B4 is also associated with minor U12-dependent spliceosome, which is involved in the splicing of rare class of nuclear pre-mRNA intron. Defects in SF3B4 have been linked to AFD1 (acrofacial dysostosis type 1) which is a form of acrofacial dysostosis, a group of disorders characterized by malformation of the craniofacial skeleton and limbs.
