RBM10 Overexpression Lysate

HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.

The protein encoded by this gene contains RNA recognition motif found in a variety of RNA binding proteins, including various hnRNP proteins, proteins implicated in regulation of alternative splicing, and protein components of snRNPs. In vitro studies showed that the rat homolog bound to RNA homopolymers, with a preference for G and U polyribonucleotides. This gene is part of a gene cluster on chromosome Xp11.23, and its 3' end lies within 20 kb upstream of UBE1. Two transcript variants encoding different isoforms have been identified for this gene. [provided by RefSeq]. Transcript Variant: This variant (2) lacks a coding exon, and uses and alternate donor splice site in another coding exon, compared to variant 1. However, it maintains the same reading frame, and encodes a shorter isoform (2) compared to isoform 1.
