PARD3/Par3 Antibody Summary
Immunogen |
Synthetic peptide from amino acids 31-342 corresponding to rat PAR2 protein from the following sequence, was selected from the unique region of the mouse PAR3 protein, peptide was post-synthetically modified to achieve highest antigenicity before used for coupling to KLH using heterobifunctional cross linker for immunogen preparation. |
Epitope |

Specificity |
This antibody detects a band of approximately 48kDa PAR3 protein in samples. The antibody also labels a 58kDa bands in PAR3 samples, probably represents either an uncharacterized variant of PAR3 or a non-specific protein. Further experiments are needed to address these possibilities |

Background
Serine proteases are well known as enzymes involved in digestion of dietary proteins, blood coagulation, and homeostasis. A new role of serine proteases as signal molecule acting via protease-activated receptors (PARs) was recently uncovered. Important effects of PAR activation on leukocyte motility, cytokine production, adhesion molecule expression, and a variety of other physiological or pathophysiological functions have been described in vitro and in vivo. PAR activation is also involved in a variety of diseases conditions including neuroinflammatory and neurodegenerative process of skin, joints, and airway inflammation. Additionally Par activation is involved in innate and adaptive immunity (1). PAR receptors belong to the 7TMD family members and are expressed by 4 distinct genes (PAR1-4). PAR receptors and purinergic receptors (P2RY12 ) play important role in homostasis and thrombosis. Par3knockout mice showed nearly complete protection against carotid artery thrombosis caused by low FeCl(3) injury. Par3 and Par4 heterozygous mouse platelets showed partially decreased responses to thrombin, resembling those in PAR1 antagonist-treated human platelets (2). PAR3 is a thrombin receptor that is expressed in mouse megakaryocytes and is responsible for normal thrombin responses in mouse platelets (3).
