Background
NKX6.1 (homeobox protein Nkx-6.1) is a transcription factor that binds to specific A/T-rich sequences in promoter regions of several genes and is known for its involvement in transcriptional regulation in islet beta cells via NKX6.1-insulin promoter binding mediated control of insulin gene. Together with NKX2-2 as well as IRX3, NKX6.1 acts to restrict the generation of motor neurons to the appropriate region of the neural tube. It belongs to neuronal progenitor factors class II proteins that are induced by SHH signals and is a member of the homeodomain family of transcription factors responsible for multiple organ systems development. The pancreatic development involves many such homeodomain proteins including Hb9, Pdx1, Pax4, Nkx2.2, and importantly on Nkx6.1 and mice deficient for Nkx6.1 display a dramatic reduction in beta-cell numbers in pancreas with near-normal development of other glandular cell types. Moreover, Nkx6.1 deficient mice have significant defects involving V2 interneuron and motor neuron specification in the ventral spinal cord, thereby emphasizing an important parallel in developmental pathways leading to beta-cells and neurons.
