Background

Excitatory amino acid transporter 1 (SLC1A3), also known as sodium-dependent glutamate/aspartate transporter 1, GLAST-1, solute carrier family 1 member 3, EAAT1, and EA6, transports and regulates L-glutamate, D-aspartate, and L-aspartate. SLC1A3 ends the glutamate postsynaptic actions by removing remaining glutamate from the cleft and by reducing extracellular glutamate. SLC1A3 is a member of a family of high affinity sodium-dependent transporter molecules that regulate neurotransmitter concentrations at the excitatory glutamatergic synapses of the mammalian central nervous system. In addition, SLC1A3 may also be important for the prevention of glutamate excitotoxicity. The SLC1A3 protein is located on the cell membrane. SLC1A3 is prominently expressed in the cerebellum, frontal cortex, hippocampus and basal ganglia and is also reported to be found in heart, placenta, lung and striated muscle. The SLC1A3 protein is subject to glycosylation and phosphorylation. SLC1A3 has two known isoforms (59.5 and 55 kDa) formed by alternative splicing. Mutations in the SLC1A3 gene have been linked to episodic ataxia and hemiplegia (PMID: 16116111) while other mutations result in a form of SLC1A3 (E219D) which influences Tourette syndrome (PMID: 21233784).

  1. Can I use this antibody for isolating live cells by using FACS sorting? GLAST is a membrane transporter. So I expected that we can sort live cells using this antibody. Can we?
    • Whether the antibody can be used with live cells, that would depend on two criteria: the presence of any preservative and endotoxin levels. The NB100-1869AF488 does not contain preservatives; thus, this criterion is good for you cell sorting experiments. In terms of the presence of endotoxin, the harmful effect(s) will depend on the cell types (the resistance/tolerance to the endotoxin). For example, blood cells and primary cells should be readily activated by endotoxin, but other cell lines may not be so reactive. Since we don't know what kind of cells that will be used, it will need wet-bench tests to find out.
  2. Do you offer cat# NB100-91662 and NB100-1869AF488 labelled with Cy5 or APC ?
    • NB100-1869 is validated for Flow, both types of IHC (frozen and paraffin), and ICC/IF ... and already has several conjugates available.We could add this antibody conjugated to APC or Cy5 into our system if interested. NB100-91662 is not validated for Flow but has been validated for IHC (parrafin only IHC-P) ... and already has several conjugates available. We could add both conjugates, but if these are going to be used for Flow it would not be guaranteed.
