alpha -Smooth Muscle Actin was detected in immersion fixed paraffin-embedded sections of human colon cancer using Mouse Anti-Human/Mouse/Rat alpha -Smooth Muscle Actin Monoclonal Antibody (Novus Catalog # NBP2-33006) at 0.25ug/mL at 37 ° Celsius for 2 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9). Tissue was stained using the Alexa Fluor™ 555 Goat anti-Mouse IgG Secondary Antibody at 1:100 at 37 ° Celsius for 2 minutes. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM in TBST for 1 hour at room temperature. The primary antibody (1/1,000) and Loading control antibody (Rabbit anti-GAPDH, 1/10,000) were used in 5% BSA at room temperature for 2 hours. Goat Anti-rabbit IgG-HRP Secondary Antibody (HA1001) at 1:100,000 dilution was used for 1 hour at room temperature.
