<b>Orthogonal Strategies Validation. </b>Dual RNAscope ISH-IHC: SARS Nucleocapsid Protein Antibody [NB100-56576] - Formalin-fixed paraffin-embedded tissue sections of SARS-CoV-2 infected human lung tissue were probed for SARS-CoV-2 viral RNA (ACD anti-sense specific probe v-nCoV2019-S (848561); Fast Red chromogen, ACD [322360]). Adjacent tissue section was processed for immunohistochemistry using rabbit polyclonal anti-SARS Nucleocapsid Antibody [NB100-56576] at 15ug/mL with 1 hr incubation at 25 degrees Celsius followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody [VC003] and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to SARS-CoV-2 infected cells.

Immunohistochemistry: SARS Nucleocapsid Protein Antibody [NB100-56576] - Characteristics of 2B4 cells clonally derived from human bronchial epithelial Calu-3 cells. Finally, 2B4 cells (passage 6) were infected with SARS-CoV (MOI = 0.1) for 24, 48, and 72 hrs before being fixed with 4% paraformaldehyde for monitoring the morphological changes of infected cells, as visualized by the expression of SARS-CoV NP protein (red) by using the standard IHC (F). Western Blot: SARS Nucleocapsid Protein Antibody [NB100-56576] - Western blot shows recombinant SARS-CoV-2 Nucleocapsid protein. PVDF membrane was probed with 1 ug/mL of Rabbit Anti-SARS-CoV-2 Nucleocapsid Polyclonal Antibody (Catalog # NB100-56576) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (HAF008). A specific band was detected for SARS-CoV-2 Nucleocapsid at approximately 55 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1

Western Blot: IL-1 beta/IL-1F2 Antibody [NB600-633] - Antibody was used at a 1:200 dilution incubated 1h at room temperature to detect dog IL-1b by Western blot. Lane 1 and 2 were loaded with 2.5 ug and 500 ng of dog IL-1b respectively. The molecular weight of the detected band is estimated by comparison to molecular weight markers (not shown). Detection occurred using a 1:3,000 dilution of IRDye (TM) 800 conjugated Donkey anti-Rabbit IgG for 1h at room temperature.

Immunohistochemistry: TLR7 Antibody [NBP2-24906] - Expression of the TLRs 5 and 7 were evaluated in the PDAC tissue by their staining intensity using TLR5 antibody (NBP2-24787) and TLR7 antibody (NBP2-24906). Note negative staining in E and G; positive staining in F and H. For TLR7 immunopositivity was detectable in the cytoplasm with no notable membranous or nuclear positivity. TLR5 showed distinctive nuclear positivity, and detectable positivity in the cytoplasm in some samples. This antibody will recognize 10% of the non-denatured (native) precursor 31,000 MW mouse IL-1beta containing samples but will primarily detect all of the 17,000 MW mature molecule. However, in western blot analysis, the usual procedure of heating the sample in SDS with or without reducing agents will facilitate denaturing of the 31,000 MW IL- 1beta precursor molecule. Denatured IL-1beta will have a 18 kDa band.

Immunohistochemistry-Paraffin: TLR7 Antibody [NB100-56588] - Analysis of FFPE human brain using TLR7 antibody at 1:20 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Neuronal staining of TLR7 was observed. Staining was performed by Histowiz.

Flow (Intracellular): TLR7 Antibody [NB100-56588] - Analysis using the FITC conjugate of NB100-56588. Staining of TLR7 in TLR7/HEK 293 cells using this antibody at 2 ug/10^6 cells. Green: Vector/HEK 293 cells. Red: TLR7/HEK 293 cells.
