![Immunohistochemistry-Paraffin: CCR7 Antibody [NB100-712] - M1 and M2 macrophages showed similar expression patterns in cortical bone. IHC staining was performed in sagittal serial sections of medial condyles from 6-week-old rats within the diaphyseal region. Cluster of differentiation 68 (CD68) was selected as a pan marker for the macrophage lineage, including monocytes, macrophages, giant cells, and osteoclasts. iNOS and C-C chemokine receptor type 7 (CCR7) were selected as M1 phenotypic markers, whereas Arginase1 and cluster of differentiation 163 (CD163) were selected as M2 phenotypic markers. Tartrate-resistant acid phosphatase (TRAP) staining was performed as described in Materials and Methods. Quantification of cell numbers according to their location revealed that most of the M1 and M2 cells were not attached to the bone surfaces. The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2. ![Immunohistochemistry-Paraffin: CXCR3 Antibody [NB100-56404] - IHC analysis of a FFPE human spleen using 1:25 dilution of CXCR3 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of HIER using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (40X) was performed using Aperio AT2 (Leica Biosystems).
