The receptor tyrosine kinase TrkA binds nerve growth factor (NGF) with low affinity, initiating a signaling cascade that mediates neuronal survival and differentiation. Higher affinity binding of NGF requires a complex formed between TrkA and the p75 NGF receptor. Ligand binding induces receptor dimerization and autophosphorylation on multiple tyrosine residues, including Y490, Y751 and Y785. Phosphorylation of TrkA at Y785 creates a binding site for phospholipase C-gamma (PLC-gamma ), which catalyzes the cleavage of substrate PIP2 to generate the second messengers DAG and IP3.
