Background

Desmin is the protein subunit of muscle-type intermediate filaments (IFs). Desmin is one of the five major groups of IFs and is found predominantly in skeletal, cardiac, and smooth muscle.DE-U-10 labels desmin in tumors derived from muscle tissue (leiomyomas and rhabdomyosarcomas) (Debus E et al.).

Desmin was detected in immersion fixed frozen sections of mouse skeletal muscle using Human Desmin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3844) at 10 µg/mL overnight at 4 °C. Tissue was stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Specific staining was localized to z-lines. Myosin Heavy Chain was detected in immersion fixed paraffin-embedded sections of human skeletal muscle using Mouse Anti-Myosin Heavy Chain Monoclonal Antibody (Catalog # MAB4470) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to sarcoplasm.
