In Western blot, a band is seen at 29 kDa. Additional bands may be seen at 36 kDa representing p35srj, the alternatively spliced version of CITED2, and at 70 kDa (unknown). Does not work well on mouse tissue. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Background

CITED2 (Cbp/p300-interacting transactivator 2; also called MSG-related protein 1/ MRG1 or P35srj) is a CREBBP/EP300-interacting protein which is highly conserved in placental mammals with 95% identity shared between human and mouse protein. CITED2 is a transcriptional co-activator that stabilizes the interactions between DNA-binding proteins and the histone acetyltransferases CBP (CREB -binding protein) as well as p300. It is localized in the nucleus and can be induced upon hypoxic trigger wherein it functions as a negative regulator of HIF1 mediated signaling by competing with HIF1alpha for binding to CBP/p300. Additionally, CITED2 gets induced by several biological stimuli such as cytokines, serum and LPS in different cell type. CITED2 interacts with a diverse array of proteins including SMAD2, SMAD3, LHX1/2, ESR1/2, WT1, EP300, PPARA, TFAP2A, TFAP2B, TFAP2C, HNF4A etc and exerts versatile biological activities. CITED2 is a critical transcriptional cofactor involved in multiple organ development and mice lacking CITED2 die in utero with cardiac and aortic arch malformations, adrenal gland agenesis, small cranial and dorsal root ganglia, exencephaly, and neural crest and left-right patterning defects. Defects in CITED2 have been linked to ventricular septal defect type 2 (VSD2) and atrial septal defect type 8 (ASD8).
