This Cathepsin E antibody is useful for Immunocytochemistry/Immunofluorescence, Immunohistochemistry paraffin embedded sections and Western blot, where a single band of ~42 kDa is detected. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Background
Cathepsin E (CTSE) is an intracellular aspartic protease belonging to pepsin superfamily and is highly homologous to the analogous aspartic protease cathepsin D. CTSE is predominantly expressed in cells of immune system including antigen-presenting cells (APC) and has also been detected in gastric epithelial cells as well as osteoclasts. Intracellularly, CTSE is localized in endosomal compartments, plasma membrane, ER and Golgi complex, and is found secreted as activated phagocytes's catalytically active enzyme. CTSE expression and secretion increases in APCs when activated by interferon- gamma and/or LPS. CTSE functions in vivo at physiological pH with a rather restricted specificity and its strategic expression alongwith localization suggests its critical involvement in tissue-specific biological functions which includes antigen processing , neuronal degeneration and generation of various secretory proteins. Human subjects and mice with shortage of CTSE are often prone to atopic dermatitis. Increased CTSE is observed in proliferative diseases such as gastric carcinoma, cervical cancer, and has been considered a promising marker for pancreatic duct cancer.
