Application Notes
This Adenylate Cyclase 3 antibody is useful for Immunocytochemistry/Immunofluorescence and Western blot, where bands can be seen at 160-200 kDa (depending on the tissue). In WB, bands can be seen at about 200kDa in olfactory epithelium which is rich in cilia. Fewer cilia are found in frontal cortex, and the protein is less heavily glycosylated, and a less prominent band is seen at about 160kDa.

Background

The biosynthesis of cAMP/cyclic 3'5'-AMP from ATP is catalyzed by adenylyl cyclases (ADCYs or ACs) which are a groups of enzymes having about ten closely related isoforms including ADCYs 1-9 and ADCYAP1 (ADCY activating polypeptide 1). Adenylate Cyclase 3 (ADCY3 or AC3) is third member of ADCYs family which is a membrane-associated enzyme and catalyzes the formation of secondary messenger cAMP. ADCY3 enzyme is localized in the membranes surrounding cilia in neurons, and is considered an excellent marker of neuronal cilia in brain tissues or in cell culture. Human ADCY3 is a large complex molecule of 1145 amino acids with an expected molecular weight of 129kD and due to extensive glycosylation, a diffused band of about 160kD in cortex and about 200kD in olfactory epithelium is generally observed in Western blots. ADCY3 appears to be widely expressed in various human tissues wherein it implicates in a number of physiological (including odorant detection via controlling intracellular cAMP levels) as well as pathophysiological metabolic processes. ADCY3 knock out mice exhibits obesity upon ageing primarily because of low locomoter activity, hyperphagia and leptin insensitivity. Moreover, ADCY3 genetic polymorphisms have been linked to decreased risk of body mass index/BMI among the subjects with obesity and type 2 diabetes/NIDDM suggesting that ADCY3 is a novel candidate in the regulation of body weight.
