Dual RNAscope ISH-IHC: xCT Antibody [NB300-318] - Formalin-fixed paraffin-embedded tissue sections of human stomach were probed for xCT mRNA (ACD RNAScope Probe, catalog #422688; Fast Red chromogen, ACD catalog # 322750). Adjacent tissue section was processed for immunohistochemistry using Rabbit Polyclonal (Novus Biologicals catalog # NB300-318) at 0.25ug/mL with 1 hour incubation at room temperature followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody (Catalog # VC003) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to glandular cells.

<b>Biological Strategies Validation and Orthogonal Strategies Validation.</b>Simple Western: xCT Antibody [NB300-318] - Simple Western lane view shows a specific band for xCT using NB300-318 at 25 ug/ml in HeLa and HeLa + DEM cell lysates. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

Western Blot: xCT Antibody [NB300-318] - Western blot analysis of protein isolated from subcutaneous tumors derived from in vivo growth of the clones relative to WT-derived tumors revealed that xCT levels remained low, phospho-STAT5 (p-STAT5) levels remained high, and phospho-STAT3 (p-STAT3) levels remained unchanged in the absence of SH-4-54. <b>Biological Strategies Validation. </b>Western Blot: xCT Antibody [NB300-318] - Total protein from Human HeLa cells treated with and without 0.1 mM Diethyl Maleate for 24 hours was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-xCT in 1% non-fat milk in TBST and detected with an anti-rabbit HRP secondary antibody using chemiluminescence. Note the increase in xCT expression with treatment.

Flow Cytometry: xCT Antibody [NB300-318] - An intracellular stain was performed on HeLa with xCT Antibody NB300-318 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. WNT11 is induced by hypoxia or hypoxic mimetics in different cell types.(A) Increased Wnt11 mRNA in EMSC adipocytes (Day 12) after hypoxia-mimetic treatments. Effect of PGE1 on stability and synthesis of HIF-1 alpha .(A) Human aortic smooth muscle cells (HASMCs) and human umbilical vein endothelial cells (HUVECs) were exposed to 1 µM PGE1 or 100 µM DFX or incubated for 4 h, and CHX was added to a final concentration of 100 µM. The cells were incubated for 0 to 30 min, and whole-cell lysates were subjected to immunoblot assay using anti-HIF-1 alpha or -beta antibodies. (B) Serum-starved HASMCs were pretreated with no drug and 1 µM PGE1 for 30 min in Met-free medium. [35S]Met-Cys was added, and the cells were incubated for 60 min prior to preparation of cell lysates. Aliquots of 1 mg of the lysates were subjected to immunoprecipitation with anti-HIF-1 alpha antibody, separated by SDS-PAGE and exposed. Aliquots of 50 µg of the same lysate were separately subjected to immunoblotting analysis with anti-beta -actin antibody. Zebrafish Vwf forms pseudo-Weibel-Palade bodies (pseudo-WPBs) in mammalian cell culture. pVWF/Myc-HIS (human VWF, (a–c)) or pzVwf/Myc-HIS (zebrafish Vwf, (d–i)) plasmids were transfected into HEK293T cells. Knockdown Validated: UNG Antibody [NBP1-49985] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and UNG knockout (KO) HeLa cell line. PVDF membrane was probed with 1:2000 of Rabbit Anti-Human UNG Polyclonal Antibody (Catalog # NBP1-49985) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for UNG at approximately 38 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

<b>Genetic Strategies Validation. </b>Western Blot: UNG Antibody [NBP1-49985] - Detection of Human UNG by Western Blot and Immunoprecipitation. Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) and 293T (T; 50 mcg) cells.
