Background
ZMPSTE24 (zinc metalloproteinase Ste24 homolog) is a zinc metalloprotease that catalyzes the removal of C-terminal three residues of farnesylated proteins and is essential for maturation of lamin A (a nuclear scaffold protein). ZMPSTE24 belongs to peptidase M48A family and is a membrane-associated enzyme which contains 7-transmembrane segments localized to ER membrane as well as inner nuclear membrane. ZMPSTE24 contains a consensus zinc metalloprotease motif, consisting of a HEXXH catalytic site, located in a loop predicted to reside in the cytosol/nucleoplasm and farnesylated substrates such as prelamin A are accessible to the HEXXH domain for proteolytic cleavage. Lamin A, together with lamin C and B-type lamins, form the meshwork that underlies the nuclear envelope, and provide structure/shape to the nucleus along with playing many critical roles in nuclear function such as binding of heterochromatin, regulation of transcription factors, positioning of nuclear pore complexes and facilitating response to DNA damage. Defects in ZMPSTE24 have been linked to mandibuloacral dysplasia with type B lipodystrophy (MADB) and lethal tight skin contracture syndrome (LTSCS).
