Rap2A and Rap2B are members of the small GTPase superfamily and ras family of proteins. Both proteins are 183 amino acids (aa) in length (SwissProt P10114 and P61225), and they are 90% identical at the amino acid level, with the most variability at the carboxyl terminus of the protein. Unlike normal Ras proteins that have a nontransforming glutamine at aa 61, these two oncogenic proteins have a threonine in that position. This change makes the intrinsic GTPase activity for these proteins lower, thereby allowing them to exist in the activated state for a longer period of time than normal Ras proteins.
