Background
Pin1 (peptidyl-prolyl cis-trans isomerase NIMA-interacting 1) is a small enzyme which is unique amongst all peptidyl-prolyl isomerases (PPIases) as it interacts with phosphorylated serine/threonine residues followed by a proline (pS/T-P). Localized in nucleus and cytoplasm, Pin1 interacts with STIL, KIF20B, NEK6, PRKX, BTK, DAPK1, ATCAY and phosphorylated RAF1. It specifically isomerizes pS/T-P peptide bonds of its target proteins and considering the number of phosphoproteins with pS/T-P motifs in cells, it is not surprising that Pin1 has such profound influence on multiple biological processes. ATM/ATR facilitates its phosphorylation upon DNA damage and DAPK1 mediated phosphorylation at Ser-71 leads to inhibition of its catalytic activity, nuclear localization, and its ability to induce centrosome amplification, chromosome instability and cell transformation. Pin1 is regarded as regulatory timer of cell signaling processes and it is implicated in transcription and enzymatic activity, protein phosphorylation, stability, processing, localization and interaction, cellular growth/proliferation, apoptosis and immune responses. Moreover, Pin1's deregulation contributes to many human pathologies such as aging, cancer, neurological disorders including Alzheimer's disease, autoimmune and inflammatory diseases.
