In Western blot analysis, a band is seen at ~16 kDa representing p14ARF (p14ARF tends to run slightly higher than the theoretical MW of 14 kDa). Additional non-specific bands may be seen at ~25 and 47 kDa. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. |

Background
ARF (alternative reading frame; p14ARF in human, p19ARF in mouse) is a tumor suppressor that serves as a sensor of hyper-proliferative signals, resulting in p53-dependent growth arrest and apoptotic cell death. ARF interacts with several proteins including COMMD1, CDKN2AIP, TBRG1, E4F1, C1QBP etc. and can bind to BCL6, E2F1, HUWE1, MDM2, MYC, NPM1/B23, TOP1/TOPOI as well as UBE2I/UBC9. ARF has the capability of inducing cell cycle arrest in G1 as well as G2 phases. While ARF is not expressed at significant levels in most normal tissues, oncogene activation triggers ARF overexpression, resulting in inhibition of MDM2 ubiquitin ligase and stabilization of p53. It is induced by progesterone as well as Ras/DMTF1, and is downregulated through NFkB p65 subunit (RELA). Inhibition of the p53 pathway, most commonly via mutations in p53 itself, inactivation of ARF, or amplification of MDM2 has been proposed to be a crucial step in pathogenesis of most human malignancies. Moreover, mice with null ARF have been shown to develop tumors rapidly and to display delayed mammary gland involution.

While looking for antibodies targeting human p14ARF, I found a question: Based on the information on UniProt, human p14ARF has the length of only 132 a.a. However, the immunogen of NB200-159 is described as ... between residue 125 and the C-terminus (residue 173)..., ...between residues 100-173... for NB110-59085, and ...between residues 50-150... for NB200-111.
- Since these antibodies were produced, UniProt has since truncated the protein sequence. These antibodies are still directed towards p14ARF and the immunogens will be updated on our website to the updated amino acid ranges. Since you won't be able to see this until this weekend or early next week, the immunogen for NB200-111 falls between amino acids 50 and 132. The immunogen for NB200-159 falls between amino acids 90 and 132 and the immunogen for NB110-59085 falls between amino acids 50 and 132.
