This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Goat Serum as well as purified and partially purified Fructose-6-Phosphate Kinase [Rabbit muscle]
Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use. |

Application Notes |
This product has been tested for use in ELISA, immunofluorescence microscopy and western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 48 kDa in size corresponding to the processed mature form of F6PK protein by western blotting in the appropriate cell lysate or extract. |
Reactivity Notes
Cross reactivity against Muscle Phosphofructokinase/PFKM/PFK-1 from other sources is expected based on high degrees of sequence homology for muscle derived F6PK. Partial reactivity may occur against F6PK isolated from liver.

Background
Phosphofructokinases (PFK) are regulatory glycolytic enzymes that convert fructose 6-phosphate and ATP into fructose 1,6-bisphosphate (through PFK-1), fructose 2,6-bisphosphate (through PFK-2) and ADP. Human PFK-1 is tetrameric and isoenzymes include PFK-1 muscle (PFKM, PFK-A), PFK-1 liver (PFKL, PFK-B) and PFK-1 platelet (PFKP, PFK-C, PFKF). PFK-1 is inhibited by ATP and citrate (from the tricarboxylic acid cycle). PFK-1 undergoes activation in the presence of elevated AMP. The most potent activator is fructose 2,6-bisphosphate, which is produced by PFK-2 from the same substrate, fructose 6-phosphate. PFK-2 is bifunctional and a key regulator for PFK-1. PFK-2 catalyzes the synthesis of fructose 2,6-bisphosphate and contains fructose 2,6-biphosphatase activity that catalyzes the degradation of fructose 2,6-bisphosphate. PFK-2 is dimeric and isoenzymes include PFK-2 liver (PFKFB1, PFRX), PFK-2 cardiac (PFKFB2), PFK-2 placental (PFKFB3, inducible PFK-2) and PFK-2 testis (PFKFB4).
