IL-22BP/IL22 RA2 Overexpression Lysate

HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.

The protein encoded by this gene is a soluble class II cytokine receptor. This protein has been shown to specifically bind to interleukin 22 (IL22), block the interaction of IL22 with its cell surface receptor, and thus inhibit IL22 activity. This protein functions as an IL22 antagonist, and may be important in the regulation of inflammatory response. Three alternatively spliced transcript variants encoding distinct isoforms have been described. [provided by RefSeq]. Transcript Variant: This variant (2) lacks a segment within the coding region, which leads to a translation frameshift. The resulting isoform (2) has a distinct and shorter C-terminus, as compare to isform 1.
