CDC2/CDK1 Antibody (CDK1/873) - Azide and BSA Free Summary

Description
1.0 mg/ml of antibody purified from Bioreactor Concentrate by Protein A/G. Prepared in 10mM PBS WITHOUT BSA & azide. Also available at 200 ug/ml WITH BSA & azide (NBP2-44915).

Antibody with azide - store at 2 to 8C. Antibody without azide - store at -20 to -80C. Antibody is stable for 24 months. Non-hazardous.
Immunogen
Recombinant human full-length CDC2/CDK1 protein. (Uniprot: P06493)
Localization
Nuclear and Cytoplasmic
Specificity
Recognizes a 34kDa protein, which is identified as cyclin dependent kinase 1 (cdk1) or p34cdc2 protein kinase. cdk1 / p34cdc2 plays a crucial role during cell division and is most active during mitosis. It is predominantly localized in the nucleus. It is a serine/threonine kinase, which is activated by cyclin, presumably by de-phosphorylation of tyrosine residues. Activated cdk1 / p34cdc2 performs specific functions during mitosis, including nuclear envelope breakdown and chromosome condensation.

Clonality
Monoclonal

Purity

Applications/Dilutions

Dilutions
  • Immunohistochemistry
  • Immunohistochemistry-Paraffin 2 - 4 ug/ml
  • Western Blot
Application Notes
Immunohistochemistry (Formalin-fixed): 2-4ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Theoretical MW
34 kDa.
Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Reactivity Notes

Does not react with Mouse, Rat, Xenopus, drosophila, S, pombe or S. cerevisiae.

Background

The cell division control protein cdc2, also known as cyclin-dependent kinase 1 (Cdk1) or p34/cdk1, plays a key role in the control of the eukaryotic cell cycle, where it is required for entry into S-phase and mitosis. Cdc2 exists as a complex with both cyclin A and cyclin B. The best characterized of these associations is the Cdc2 p34 cyclin B complex, which is required for the G2 to M phase transition. Activation of Cdc2 is controlled at several steps including cyclin binding and phosphorylation of threonine 161. However, the critical regulatory step in activating cdc2 during progression into mitosis appears to be dephosphorylation of Tyr15 and Tyr14. Phosphorylation at Tyr15 and inhibition of Cdc2 is carried out by WEE1 and MIK protein kinases while Tyr15 dephosphorylation and activation of Cdc2 is carried out by the cdc25 phosphatase. The isoform CDC2deltaT is found in breast cancer tissues. Furthermore, cdc2/Cdk1 is a key mediator of neuronal cell death in brain development and degeneration.
