Immunocytochemistry/Immunofluorescence: Apolipoprotein A-I/ApoA1 Antibody - BSA Free [NBP2-52979] - HepG2 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with Apolipoprotein A-1/ApoA1 Antibody (NBP2-52979) at 1ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes.  Nuclei were counterstained with DAPI (Blue).  Cells were imaged using a 100X objective and digitally deconvolved.Western Blot: Apolipoprotein A-I/ApoA1 Antibody [NBP2-52979] - Western blot analysis using Apolipoprotein A-I/ApoA1 antibody. Western Blot: MDR1/ABCB1 Antibody (SN06-42) [NBP2-67667] - Analysis of P Glycoprotein on mouse heart tissue lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody  at 1:200,000 dilution was used for 1 hour at room temperature.Immunohistochemistry-Paraffin: MDR1/ABCB1 Antibody (SN06-42) [NBP2-67667] - Immunohistochemical analysis of paraffin-embedded mouse brain tissue using anti-MDR1/ABCB1 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Formalin-fixed paraffin-embedded tissue sections of human prostate cancer were probed for ABCA1 mRNA (ACD RNAScope Probe, catalog # 432291; Fast Red chromogen, ACD catalog # 322360). Adjacent tissue section was processed for immunohistochemistry using rabbit polyclonal  (Novus Biologicals catalog # NB400-105) at 1.5ug/mL with overnight incubation at 4 degrees Celsius followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody (Catalog # VC003) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to glandular cells.HSKMCs were transfected with scrambled siRNA (scr siRNA) or ABCA1 siRNA. The cells were lysed and ABCA1 (A) protein levels were quantified by immunoblotting. Western Blot: ABCG1 Antibody - BSA Free [NB400-132] - ABCG-1 protein level in control and glycolaldehyde-treated macrophages. J774 macrophages were overloaded with oxidized LDL (50 ug/mL de DMEM) for 48 h. In the last 5h, cells were incubated with 0.25 mM glycolaldehyde (GAD; black bars) and 1 uM LXR agonist (T0901317). Control cells (C; gray bars) were incubated with T0901317 alone. Cells were scrapped into TBS containing protease inhibitors. Equal amounts of cell protein were applied into a 6% polyacrylamide electrophoresis gel and immunoblot was performed utilizing anti ABCG-1 antibody.
