NB100-1761
Western blot shows lysates of HAP1 human near-haploid cell line and Matrin 3 knockout HAP1 cell line (KO). Nitrocellulose membrane was probed with Matrin 3 Antibody (Catalog # NB100-1761) O/N at 4C, followed by HRP-conjugated Secondary Antibody and ECL detection. A specific band was detected for Matrin 3 (as indicated) in the parental HAP1 cell line, but is not detectable in knockout HAP1 cell line. Primary antibody dilution used: 1/5000. The Ponceau stained transfers of each blot are shown. Image, protocol and testing courtesy of YCharOS Inc. (ycharos.com).HAP1 lysates were prepared, and immunoprecipitation was performed using 2.0 ug of Matrin 3 antibody (Catalog # NBP1-04340) pre-coupled to Dynabeads protein A. Samples were washed and processed for Western Blot with Matrin 3 antibody. For Western Blot, Rb x Matrin 3 was used at 1/500. The Ponceau stained transfers of each blot are shown. SM=4% starting material; UB=4% unbound fraction; IP=immunoprecipitate. Image, protocol and testing courtesy of YCharOS Inc. (ycharos.com).

Western Blot: Tau Antibody (2E9) [NBP2-25162] - Analysis of different tissue lysates using mouse mAb to MAP-tau, NBP2-25162, dilution 1:2,000 in green: [1] protein standard (red), [2] rat brain, [3] rat spinal cord, [4] mouse brain, [5] mouse spinal cord. Tau protein is expressed as up to 9 different isoforms of different molecular weight, and so appears as multiple closely spaced bands in the range from 48 kDa to 67 kDa in the CNS and including larger big tau forms in the PNS, visible in lane 5.Immunocytochemistry/Immunofluorescence: Tau Antibody (2E9) [NBP2-25162] - Analysis of cortical neuron-glial culture from E20 rat stained with mouse mAb to MAP-tau, NBP2-25162, dilution 1:1,000 in green, and costained with chicken pAb to MAP2, dilution 1:5,000 in red. The blue is DAPI staining of nuclear DNA. NBP2-25162 antibody stains perikarya, dendrites and axons of neurons, while MAP2 antibody labels only dendrites and perikarya. As a result, perikarya and dendrites appear orange-yellow, since they contain both proteins.

NB100-1558
Western Blot: p97/VCP Antibody [NB100-1558] - Zymographic assay of gelatinase activity. Medium supernatants of 20ml culture medium (medium control) as well as 20 ml conditioned medium from 107 U937 cells, pMTH1-U937 cells and asCD11b-U937 cells in the absence or presence of 5nM TPA for 72 h, respectively, were 18-fold concentrated and subjected to SDS-PAGE containing 2 mg/ml of gelatine. Following incubation with MMP enzyme buffer, the gels were stained with 0.4% Coomassie blue and afterwards destained again to visualize the appearance of gelatinase activity by light bands against the dark background. The molecular weight markers on both sides of the gels indicate the size of the MMPs exhibiting gelatinase activities.
