Pre-treatment with 0.5 mM sodium arsenite (SA) enhances permissivity in a cell-type-specific manner across reovirus strains. EGFR was used as a membrane marker and GAPDH as a cytosolic marker. SDS-PAGE western blot validation of the increased abundance of Annexin V, Na/K ATPase and alpha-tubulin in the EVs derived from TIS Cal51 cells compared with EVs derived from non-senescent non-treated controls. EVs isolated from TIS and control cells were immunoblotted to validate the protein expression of annexin V, Na/K ATPase and alpha-tubulin from the MS data. STA-ET-8.2 cells were FACS-sorted and CD133+ and CD133- fractions treated with increasing concentrations of (A) Doxorubicin (Doxo), (B) Etoposide (Etop), (C) Vincristine (Vinc) or (D) a combination of all three drugs at high (Doxo 10 uM; Etop 10 ug/ml; Vinc 100 ng/ml), medium (Doxo 2.5 uM; 20 Etop 2.5 ug/ml; Vinc 25 ng/ml) and low (Doxo 0.5 uM; Etop 0.5 ug/ml; Vinc5 ng/ml) concentrations. Viability was assessed by MTS assay after 96 hrs (*p < 0.05) and CD133+ cells displayed increased drug resistance. (E): CD133+ and CD133- TC-71 cells are equally sensitive to Doxo, Etop, and Vinc. (F): Western blot reveals no difference in apoptosis and drug resistance protein expression between unsorted and CD133-sorted STA-ET-8.2 cell fractions. Meiotic spreads show deletion of Dicer1 disrupts progression of meiosis I.(A) It was possible to identify the 5 sub-stages of meiosis I by combining the reactivity patterns of two antibodies against synaptonemal complex protein 3 (SYCP3) and gammaH2AX. SYCP3 is a protein essential for synapsis of homologous chromosomes and gammaH2AX localizes to double-strand breaks and XY bodies during meiosis. A total of 200 spreads were counted in testis cell preparations from WT and GCKO mice at P22. Western Blot: GAP-43 Antibody [NBP1-41123] - Total protein from Human, Mouse and Rat brain, and SHSY-5Y cells was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-GAP43 in 1% non-fat milk in TBST and detected with an anti-sheep HRP secondary antibody using chemiluminescence.
