Analysis of GAPDH in human nucleus pulposus cell culture (40ug/lane) using anti-GAPDH antibody. LI-COR Odyssey imaging system was used for detection. Western blot image submitted by a verified customer review.

Analysis of of HeLa cells stained with GAPDH antibody, dilution 1:100 (Green). DAPI staining of nuclear DNA (Blue). The GAPDH antibody produces strong cytoplasmic staining of healthy cells.

Analysis of cell line lysates probed with mouse mAb to GAPDH (theoretical molecular weight: 36 kDa), NB300-221, dilution 1:2,000: [1] protein standard, [2] HEK293, [3] HeLa, [4] SH-SY5Y, [5] COS1, [6] NIH-3T3, and [7] C6 cells. The GAPDH antibody reveals a single band at ~37 kDa in all cell lines. GAPDH is a house keeping protein, the level of which is relatively unaffected by most experimental manipulations, and, as a result, this antibody has been widely used as a western blotting loading control.

Immunohistological analysis of brain stem section stained with rabbit polyclonal Laminin Antibody [NB300-144], dilution 1:1,000 in red, and costained with chicken pAb to Myelin Basic Protein (MBP), dilution 1:5,000 in green. The blue is DAPI staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45 uM, and free-floating sections were stained with the above antibodies. The laminin antibody is an excellent marker of basement membranes surrounding blood vessels, while the MBP antibody stains the myelin sheathes around axons.

MxIF analysis of mouse submandibular salivary gland morphogenesis. Staining of rat spinal cord and dorsal root paraformaldehyde/paraffin-embedded tissue using Laminin Antibody [NB300-144]. Pepsin antigen retrieval was performed on this tissue sample.

Analysis of rat heart cells lysates (lane 1) and 0.2 ug of purified laminin protein from mouse EHS sarcoma (lane 2) Laminin Antibody [NB300-144] recognizes 3 laminin isotypes: alpha 1 (440 kDa), beta 1 (220 kDa) and gamma 1 (220 kDa). Also recognized is a laminin binding protein at 120 kDa in both rat heart lysates and purified laminin protein. Since this protein always coexpresses with laminin this crossreactivity is irrelevant. Mouse primary cultures stained with MAP2 antibody. ICC/IF image submitted by a verified customer review.

Immunofluorescent analysis of cortical neuron-glial cell culture from E20 rat stained with chicken pAb to microtubule associated protein 2 (MAP2), NB300-213, dilution 1:10,000 in red, and mouse mAb to MAP-tau, NPB2-25162, dilution 1:2,000, in green. The blue is DAPI staining of nuclear DNA. NB300-213 antibody stains dendrites and perikarya of neurons, while NBP2-25162 antibody labels neuronal perikarya, dendrites and also axonal process. As a result perikarya and dendrites appears orange-yellow, since they contain both MAP2 and tau, while axons are green.

Western blot analysis of whole brain tissue lysates using chicken pAb to microtubule associated protein 2 (MAP2), NB300-213, dilution 1:50,000 in green: [1] protein standard (red), [2] adult rat brain, [3] embryonic E20 rat brain, [4] adult mouse brain. Strong band observed at ~280 kDa mark corresponds to two major isoforms of MAP2 protein referred to as MAP2A and MAP2B. Smaller fragments of these isoforms are also detected if the antibody is used at higher concentrations.

PEA decreases astrocyte activation in organotypic cultures of rat hippocampi and rescues neuronal CA3 damage caused by Abeta challenge. Abeta-challenged (1 ug/ml) slices of rat hippocampi were treated for 24 hours with PEA (0.1 uM) in the presence of the selective PPAR? antagonist (GW9662, 9 nM) or the selective PPARalpha antagonist (MK886, 3 uM). PEA inhibits astroglial proliferation and reduces neuronal loss in mixed neuroglia co-cultures exposed to Abeta. Abeta-challenged (1 ug/ml) astrocyte/neuron mixed cultures were treated with PEA (0.1 uM) i9662, 9 nM) on the presence of the selective PPAR-gamma antagonist (GWr the selective PPARalpha antagonist (MK886, 3 uM). Immunohistochemistry: GFAP Antibody [NB300-141] - Analysis of a rat cerebellum section stained with rabbit polyclonal antibody to GFAP, NB300-141, dilution 1:5000 in green and mouse monoclonal antibody to MeCP2, dilution 1:500, in red. The blue is DAPI staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 1 hour, cut to 45 uM, and free-floating sections were stained with above antibodies. The GFAP antibody stains the network of astrocytic cells and the processes of Bergmann glia in the molecular layer. The MeCP2 antibody specifically labels nuclei of certain neurons.

Increased astrocyte activation in IL-1beta-stimulated two-week old offspring is reduced by maternal LB supplementation. Western Blot: mCherry Antibody (1C51) [NBP1-96752] - Analysis of HEK293 cell lysates and recombinant protein solutions using mCherry antibody, dilution 1:1,000 (Green). [1] protein standard, [2] HEK293, [3] HEK293 cells transfected with mCherry-HA construct, [4] mCherry recombinant protein, [5] GFP recombinant protein, and [6] HEK293 transfected with GFP construct. Major band at about 30 kDa corresponds to mCherry protein (predicted molecular weight: 27 kDa). mCherry antibody does not react with GFP protein. The same blot was simultaneously probed with chicken HSP60 pAb, dilution 1:5,000 in red which reveals band at 60 kDa seen only in cell lysates.

Mutation of threonine 276 to aspartic acid promotes the association of cytohesin 2 with membranes. MDCK cells were transfected with constructs encoding mCherry-tagged wild-type or T276D cytohesin 2 and fractionated into cytosol and total membranes. Western Blot: mCherry Antibody (1C51) [NBP1-96752] - WB assay of the crude extract of HEK293 cells transfected with pFin-EF1-mCherry vector (lane +) and an equal amount of protein extract from untransfected HEK293 cells (lane -). NBP1-96752 binds a major band running at about 28 kDa (observed molecular weight) corresponding to intact full-length mCherry. The two other bands are clearly processed forms of mCherry as they are not present in non-transfected HEK293 cells.

Immunocytochemistry/Immunofluorescence: mCherry Antibody [NBP2-25157] - HEK293 cells transfected in the same way and viewed in the confocal microscope. Most HEK293 cells are not transfected so only the nucleus of these cells can be visualized with a blue DNA stain. Cells which are transfected with mCherry are red. Staining with NBP2-25157 is shown in Green. Green antibody staining is only seen in cells which express mCherry, as expected, and the superimposition of the green and red signals results in an orange signal. Interestingly, stronger mCherry staining is seen in the nucleus, possibly due to degradation of some mCherry molecules to release the low molecular weight mCherry fluorochrome. Western Blot: Vimentin Antibody [NB300-223] - Analysis of tissue and cell lysates. Antibody at 1:5000 in red. [1] protein standard (red), [2] rat whole brain lysate, [3] HeLa, [4] SH-SY5Y, [5] HEK293, and [6] NIH-3T3 cell lysates. NB300-223 binds to the vimentin protein showing a single band at ~50 kDa. The blot was simultaneously probed with mouse mAb to MAP2C/D, dilution 1:5000 in green, revealing multiple bands around 280 kDa that correspond to full length MAP2A/2B isotypes, and ~70 kDa bands which are MAP2C/D isotypes. MAP2 isotypes are seen only in extracts containing neuronal lineage cells.

Formalin fixed rat brain stem neurons stained with to NeuN in green and counter stained with , our chicken polyclonal antibody to microtubule associated protein 2 in red. The nuclei of cells are revealed with DAPI in blue. The antibody reveals strong nuclear and distal cytoplasmic staining for Fox3/NeuN and the complete absence of staining of other cell types. The MAP2 antibody binds to dendrites and overlaps with Fox3 staining in perikarya. This Fox3/NeuN antibody is therefore an excellent marker of neuronal cells.

Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 700.

Rat brain neural cultures stained with NBP1-92693 (red), a chicken polyclonal antibody to GFAP (NBP1-05198, green) and DAPI (blue). NBP1-92693 reveals strong nuclear and distal cytoplasmic staining for RBFOX/Neun and the complete absence of staining of astrocytes, which are staining with the GFAP antibody, and other kinds of non-neuronal cells. This RBFOX/Neun (1B7) antibody is therefore an excellent marker of neuronal cells.

IHC-Fr analysis of a 4% PFA fixed mouse spinal cord tissue section using RBFOX3/NeuN antibody (clone 1B7) at 1:500 dilution with overnight 4C incubation in a dilution buffer which contained BSA 1% and 0.2% TritonX100 in PBS. The signal was detected using Alexa Fluor 594 conjugated goat anti-mouse IgG (H+L) secondary antibody. The antibody generated a specific cytoplasmic-nuclear signal and the staining was more intense in the nuclei of the neurons. Immunocytochemistry/Immunofluorescence: mCherry Antibody [NBP2-25158] - HEK293 cells transfected with mCherry, stained with mCherry antibody and viewed in a confocal microscope. Most HEK293 cells are not transfected so only the nucleus of these cells can be visualized with a blue DNA stain. Cells which are transfected with mCherry are bright red, and staining with the mCherry antibody is shown in Green. The green antibody staining is only seen cells which express mCherry, and the superimposition of the green and red results in an orange signal. Interestingly stronger mCherry staining is seen in the nucleus, possibly due to degradation of some mCherry molecules releasing the low molecular weight mCherry fluorochrome.

Western Blot: mCherry Antibody [NBP2-25158] - Analysis of NBP2-25158. Lane 1. Lysate of HEK293 cells transfected with pFin-EF1-mCherry vector. There is a strong clean band at about 29kDa corresponding to mCherry (predicted molecular weight ~27kDa). Lane 2, lysate of untransfected HEK293 cells show no protein bands. The NBP2-25158 antibody was used at 1:2,000 dilution.

Western Blot: mCherry Antibody [NBP2-25158] - Analysis of HEK293 cell lysates, and recombinant protein solutions using chicken mCherry pAb, dilution 1:2000 (Green). [1] protein standard, [2] HEK293, [3] HEK293 cells transfected with an mCherry-HA constract, [4] pure recombinant mCherry protein (predicted molecular weight ~ 27kDa), [5] pure recombinant GFP protein and [6] HEK293 transfected with a GFP construct. The major band at about 30kDa corresponds to mCherry-HA protein and the slightly larger recombinant form runs at about 33kDa due to presence of a His tag and other vector derived sequence. The mCherry antibody reacts strongly with both mCherry constructs and does not react with GFP protein. The blot was simultaneously probed with mouse HSP60 mAb, dilution 1:10000 (Red), which reveals a band at 60kDa only in the cell lysates.

Western blot analysis of HEK293 cell lysates using chicken pAb to mCherry, NBP2-25158, dilution 1:2,000, in green, and rabbit pAb to GAPDH, RPCA-GAPDH, dilution 1:5,000, in red: [1] protein molecular weight standard, [2] untransfected HEK293 control cells, [3] HEK293 cells transfected with pCI-Neo-mod vector expressing two tdTomato protein domains, [4] HEK293 cells transfected with pCI-Neo-mod vector expressing one mCherry-HA protein domain, and [5] HEK293 cells transfected with pCI-Neo-mod vector expressing one GFP domain. NBP2-25158 recognizes tdTomato and mCherry proteins revealing major bands at about 60kDa and 30kDa, but does not recognize GFP. Compound MI-743 causes intracellular 8-oxo-dG accumulation and DNA damage.a MGC-803, HGC-27 and GES-1 cells were treated with DMSO or 5 µM MI-743 for 48 h. Immunohistochemistry-Paraffin: Ubiquitin Antibody (Ubi-1) [NB300-130] - FFPE section of cerebral cortex of an Alzheimer patient processed with Ubiquitin antibody using HRP/DAB (Brown) and also stained with haemotoxylin (Blue). A typical flame shaped tangle is seen in a pyramidal neuron in the center and is surrounded by some dystrophic neurites, also strongly ubiquitin positive. Both are commonly seen in cortical and hippocampal Alzheimer brain sections and are typical for this disease, but are rare or absent in healthy brain.
