Background

TM2D2 is encoded by this gene contains a structural module related to that of the seven transmembrane domain G protein-coupled receptor superfamily. This protein has sequence and structural similarities to the beta-amyloid binding protein (BBP), but, unlike BBP, it does not regulate a response to beta-amyloid peptide. This protein may have regulatory roles in cell death or proliferation signal cascades. This gene has multiple alternatively spliced transcript variants which encode two different isoforms. Flow Cytometry: c-Myc Antibody (9E10) [NB600-302] - An intracellular stain was performed on U-937 cells with c-Myc Antibody (9E10) NB600-302AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Immunocytochemistry/Immunofluorescence: PCNA Antibody (PC10) [NB500-106] - HeLa cells were fixed and permeabilized for 10 minutes using cold (-20C) MeOH. The cells were incubated with anti-PCNA [PC10] at 5 ug/mL overnight at 4C and detected with an anti-mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
