Background

New Delhi metallo-beta-lactamase is an enzyme that predisposes bacteria to become antibiotic resistant. In Pakistan, multi-drug resistant bacteria are blamed for a higher than usual incidence of hospital infections. NDM1 has an interesting conformational design, in that it has a large active site which allows it to react with multiple substrates, and which changes distinctly after substrates bind to it. Therapies against NDM1 in tandem with antibiotic therapies are being postulated as the best potential defense against antibiotic resistance. However, the ability to identify NDM1 prevalence is also critical. Identification of colonized individuals by immunodetection assays could be a critical step in isolation and treatment.

  1. If I want to detect the presence of NDM-1 protein in our isolates (Acinetobacter), would you please let me know if I can used this Antibody via WB?
    • Here is the information on the immunogen for this antibody: A synthetic peptide made to a C-terminal region of the E. coli bla-NDM-1 protein (within residues 100-158). [NCBI BAJ10873] We have confirmed this antibody to work in WB, but I do not believe the lab has tested in Acinetobacter specifically. If the protein is expressed in those samples and should have high similarity with the sequence provided for the immunogen then most likely it should work well, but I cannot say for certain since we haven't directly tested.
