Background

Transcription factors containing a basic helix-loop-helix (bHLH) motif regulate expression of tissue-specific genes in a number of mammalian and insect systems. DNA-binding activity of the bHLH proteins is dependent on formation of homo- and/or heterodimers. Dominant-negative HLH proteins encoded by Id-related genes, such as ID4, also contain the HLH-dimerization domain but lack the DNA-binding basic domain. Consequently, Id proteins inhibit binding to DNA and transcriptional transactivation by heterodimerization with bHLH proteins (Pagliuca et al., 1995 [PubMed 7665172]).[supplied by OMIM]

A172 human glioblastoma cell line were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. Olig2/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human/Mouse/Rat Olig2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2418) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The p21 promoter was detected by standard PCR.
