Background
Gastric lipase (GL/LIPF or gastric triacylglycerol lipase) is an extremophilic enzyme which retains its activity in gastric juices fully at pH 2 under the presence of pepsin in stomach wherein the gastrointestinal lipolysis of dietary fat is initiated. Gastric lipase belongs to AB hydrolase superfamily/lipase family and is a secreted protein which is not inhibited by bile salts in the gastrointestinal tract, whereas pancreatic lipase (main lipase involved in fat digestion) requires a cofactor, namely colipase, to counteract the inhibitory effects of bile salts on lipase adsorption at lipid-water interfaces. Due to its acid resistant properties, it has been proposed as a candidate for the treatment of pancreatic enzyme insufficiency observed in chronic pancreatitis and cystic fibrosis, the gastrointestinal disorders which are characterized by exocrine pancreatic secretion deficiency, gastric acid secretion not being neutralized by pancreatic bicarbonate, and pH of the small intestine contents reaching as low as 2-3.
